High-density lipoprotein (HDL3)-associated α-tocopherol is taken up by HepG2 cells via the selective uptake pathway and resecreted with endogenously synthesized apo-lipoprotein B-rich lipoprotein particles

چکیده
α-Tocopherol (αTocH) is transported in association with lipoproteins in the aqueous milieu of the plasma. Although up to 50% of circulating αTocH is transported by high-density lipoproteins (HDLs), little is known about the mechanisms of uptake of HDL-associated αTocH. During the current study, human apolipoprotein (apo)E-free HDL subclass 3 (HDL $ ) labelled with ["%C]αTocH was used to investigate uptake mechanisms of HDL $ -associated αTocH by a permanent hepatoblastoma cell line (HepG2). HDL $ -associated αTocH was taken up independently of HDL $ holoparticles in excess of apoA-I comparable with the non-endocytotic delivery of cholesteryl esters to cells termed the ‘selective ’ cholesteryl ester uptake pathway. Experiments with unlabelled HDL $ demonstrated net mass transfer of αTocH toHepG2 cells. Time-dependent studies with ["%C]αTocH-

Lipoprotein lipase mediates an increase in the selective uptake of high density lipoprotein-associated cholesteryl esters by hepatic cells in culture.
In this study the effect of lipoprotein lipase (LPL) on the selective uptake of high density lipoprotein (HDL) cholesteryl esters (CE) by hepatic cells was investigated. Human HDL3 (d 1.125-1.21 g/ml) was radiolabeled with 125I in the protein moiety and with 3H in the CE moiety. LPL was prepared from bovine milk. Human hepatocytes in primary culture and human Hep3B hepatoma cells were incubated...
متن کاملSR-BI-mediated high density lipoprotein (HDL) endocytosis leads to HDL resecretion facilitating cholesterol efflux.
The high density lipoprotein (HDL) receptor, scavenger receptor class B, type I (SR-BI), mediates selective cholesteryl ester uptake from lipoproteins into liver and steroidogenic tissues but also cholesterol efflux from macrophages to HDL. Recently, we demonstrated the uptake of HDL particles in SR-BI overexpressing Chinese hamster ovarian cells (ldlA7-SRBI) using ultrasensitive microscopy. In...
متن کاملCharacterization of high density lipoprotein binding to human adipocyte plasma membranes.
Freshly isolated human adipocytes showed specific uptake of 125I-labeled human high density lipoprotein (HDL2 and HDL3), a portion of which could be released by subsequent incubation with excess unlabeled ligand. To study the mechanism of HDL binding, sucrose gradient-purified adipocyte plasma membranes were incubated with radioiodinated lipoprotein particles under equilibrium conditions in the...
متن کاملIn Vitro Effect of ?-Tocopherol, Ascorbic Acid and Lycopene on Low Density Lipoprotein Glycation
Nonenzymatic glycation of low density lipoprotein (LDL) is a reaction of glucose and other reducing sugars with apolipoprotein B100 (apo-B100) lysine residues. In diabetes, this reaction is greatly accelerated and is important in the pathogenesis of diabetic complications. The objective of this study was to investigate in vitro effects of ?-tocopherol, ascorbic acid and lycopene on LDL glycatio...
متن کاملLocalization and regulation of SR-BI in membrane rafts of HepG2 cells.
The scavenger receptor class B, type I (SR-BI) mediates cholesteryl esters (CE) selective uptake from low density lipoprotein (LDL) and high-density lipoprotein (HDL) particles. In a number of tissues expressing caveolin, SR-BI is localized in caveolae. We show using detergent-free sucrose gradients that SR-BI is found in membrane rafts devoid of caveolin-1 in the human hepatoma HepG2 cell. Per...
